INTRODUCTION.
Pleomorphic adenoma is a common benign salivary gland neoplasm characterised by neoplastic proliferation of ductal epithelial cells along with myoepithelial components, having a malignant potentiality. It is the most common type of salivary gland tumor and the most common tumor of the parotid gland. It derives its name from the architectural Pleomorphism (variable appearance) seen by light microscopy.
Pleomorphic adenoma gene-1 encodes a zinc finger protein with two (2) putative nuclear localization signals. PLAG1, which is developmentally regulated, has been shown to be consistently rearranged in pleomorphic adenomas of the salivary glands. PLAG1 is activated by the reciprocal chromosomal translocations involving cytogenetic location 8q12.1 and genomic coordinate 8:56,160,909 in a subset of salivary gland pleomorphic adenomas. Nine (9) other genes besides PLAG1 in the 8q12.1 cytogenetic location are listed BENEATH.
