INTRODUCTION.
The HMGA2 (High-Mobility Group AT-Hook-2) gene on chromosome 12q14.3 is a highly promiscuous oncogene. Rearrangements commonly truncate the protein after exon 3, conserving its DNA-binding domains while deleting its regulatory 3′ UTR. This protects the transcript from microRNA-mediated degradation and allows it to fuse with various partner genes across several benign and malignant tumors.

🧬 The Genomic Event & Fusion Protein.
Chromosomal Translocation: A reciprocal translocation occurs between chromosome 12 and chromosome 3.
Gene Fusion: This fuses exons 1–3 of the HMGA2 gene with exons 9–11 of the LPP gene.
Aberrant Function: The resulting chimeric protein combines the DNA-binding domain (AT-hooks) of HMGA2 with the transactivation domains (LIM domains) of LPP.
Oncogenic Drive: This mutant transcription factor abnormally activates target elements’ (PRDII, BAT-1), boosting proliferation and triggering lipomagenesis.
