INTRODUCTION.
Autosomal recessive disorders with obesity as a predominant feature include leptin deficiency, leptin receptor deficiency, prohormone convertase-1 deficiency, and proopiomelanocortin deficiency; associated features in these disorders include hypogonadotropic hypogonadism, hypoadrenalism, and short stature.
There are also syndromes associated with obesity such as Prader-Willi syndrome, Bardet-Biedl syndrome, and Cohen syndrome, among others.
Reviews have discussed the genetic, molecular, and environmental aspects of obesity and discussed the myriad associated complications, including hypertension, dyslipidemia, endothelial dysfunction, type-2 diabetes mellitus, and impaired glucose tolerance, acanthosis nigricans, hepatic steatosis, premature puberty, hypogonadism and polycystic ovary syndrome, obstructive sleep disorder, orthopedic complications, cholelithiasis, and pseudotumor cerebri.
There is evidence obesity is predominantly a polygenic and multifactorial trait. Genetic variation in some genes have been associated with susceptibility to obesity as a monogenic trait such as cocaine- and amphetamine-regulated transcript prepropeptide (CART) appears to have roles in reward, feeding, and stress, and it has the functional properties of an endogenous psychostimulant.
Cocaine- and amphetamine-regulated transcript prepropeptide (CART) is on cytogenetic location 5q13.2 and genomic coordinates 5:71,719,275-71,721,045.
